The researchers, who publish their work in the journal Molecular Psychiatry, say that heavy cocaine and heroin use have been shown to decrease dopamine release, but until now, such data regarding cannabis was notably absent.

Lead author Dr. Anissa Abi-Dargham, of the Columbia University Medical Center (CUMC) in New York, says the recent “widespread acceptance and use of marijuana” makes looking into the effects of cannabis on the brain extremely important.

She and her team explain that most drug addictions blunt dopamine release during the chronic phase of drug dependence, which results in poor outcomes.

To further investigate whether marijuana dependence is linked with similar effects, the researchers conducted their study in 11 adults aged 21-40 years who were heavily dependent on marijuana, and they matched them with 12 healthy controls.

The adults in the marijuana group started using it at around age 16 on average, became dependent by age 20 and had been dependent for around 7 years.

Nearly all users in the study smoked marijuana daily in the month leading up to the study, the researcher say.

‘Heavy use may negatively affect learning and behavior’

The researchers used positron emission tomography (PET) scans to track a radiotracing molecule that binds to the brain’s dopamine receptors. From this, they were able to measure the release of dopamine in the striatum, which is a brain region involved in memory, impulsive behavior and attention.

Additionally, the team was able to track dopamine release in other brain regions, including the thalamus, midbrain and globus pallidus.

During the study, the marijuana users stayed in the hospital for a week, during which they abstained from using it. This was to ensure that the scans were not measuring the drug’s effects.

Both before and after being given oral amphetamine to draw out dopamine release, the participants’ brains were scanned. The researchers used the percent change in the binding of the radiotracer as a sign of dopamine release capacity.

Results showed that, compared with the control group, the marijuana users’ striatum had lower dopamine release. There was also lower release in subregions that play a role in associative and sensorimotor learning, as well as in the globus pallidus.

Upon investigating the link between dopamine release in the striatum and cognitive performance on learning and working memory tasks, the researchers did not observe a difference in performance between the two groups.

However, they do note that among all participants, those who had lower dopamine release performed worse on both tasks.

The researchers conclude their study by noting that the lower dopamine release is linked with inattention and negative symptoms in marijuana users, and with “poorer working memory and probabilistic category learning performance” in both groups.

Published in the journal Nature Communications, the study suggests that BK channels in the suprachiasmatic nucleus (SCN) – located in the hypothalamus of the brain – regulate the “switch” between sleep and wakefulness.

BK channels are part of the ion channel family. In the brain, they are responsible for conducting electrical current, enabling communication between neurons, or brain cells.

According to study co-author Prof. Andrea Meredith – of the University of Maryland School of Medicine – and colleagues, these channels are particularly active in the SCN, which is the brain region involved in the regulation of circadian rhythms; it tells us when to go to sleep, how long to sleep for and when to get up.

Unfortunately, some people’s circadian rhythm, or body clock, is out of sync, which can lead to sleep disorders, such as insomnia, jet lag and seasonal affective disorder.

To find out if BK channels in the SCN play a role in circadian rhythm, Prof. Meredith and colleagues analyzed the SCN of mice. The sleep pattern of mice is opposite to that of humans; they sleep during the day but stay awake at night.

Mice unable to inactivate BK channel showed increased wakefulness

Some of the mice analyzed had been genetically modified so that their BK channels in the SCN could not be inactivated. In order to record activity in the BK channels of these mice – as well as the BK channel activity of normal mice – the team implanted electrodes in SCN neurons of the rodents.

Among normal mice, the team found that during the day, when the rodents sleep, the BK channels were inactive, but they were active during nighttime, when mice are normally awake.

An inactive daytime BK channel triggered higher neuronal activity in the SCN of mice, which led to sleep, but an active nighttime BK channel reduced neuronal activity, triggering wakefulness.

The team reached this conclusion after identifying lower neuronal activity in the mice that were unable to inactivate their BK channels, which they found led to increased daytime wakefulness.

The researchers note that, despite mice having an opposite sleep pattern to us, the association between neuronal activity and sleep-wake cycles is similar in both mice and humans.

According to the team, this study is the first to show that inactivation of BK channels is key for encoding the brain’s circadian rhythm, regulating sleep-wake cycles.

Previously, studies have suggested that the number of ion channels present on the surface of SCN cells is responsible for neuronal activity that regulates circadian rhythm. This latest research, however, shows that it is the activation – and inactivation – of BK channels that is responsible.

Prof. Meredith says:

“We knew that BK channels were widely important throughout the body. But now we have strong evidence that they are specifically and intrinsically involved in the wake-sleep cycle. That’s really exciting.”

The team says the findings could pave the way for the development of drugs that target circadian rhythms, which could help treat sleep disorders, jet lag and other conditions caused by body clock dysfunction.

Most studies investigating the health and well-being of children within same-sex parent families have found they fare just as well as children from families with different-sex parents.

However, the researchers of this latest study – including Dr. Henry Bos of the University of Amsterdam – note that the results of many of these studies “relied on convenience samples and/or fertility recruitment,” causing many people to question their accuracy.

Furthermore, many individuals continue to believe that children fare better when being raised by parents of different sexes, despite evidence to the contrary.

For their study, Dr. Bos and colleagues analyzed data from the National Survey of Child Health, from which they identified 95 female same-sex parent households and 95 different-sex parent households with children aged 6-17 years.

Both groups were matched for parent and child characteristics, and the families were considered stable, meaning they had no history of family instability – such as divorce or separation – since the birth or adoption of their child.

The team notes that male same-sex parent households were excluded from the analysis due to an insufficient sample size; only eight could be identified.

Child outcomes no different for children with same-sex parent families

Through a telephone interview with one parent per household, the researchers gathered information on the general health, emotional difficulties, coping behavior and learning behavior of each child.

The team also assessed levels of parental stress, the relationship quality between partners and the quality of parent-child relationships.

When it came to child outcomes, the researchers identified no differences in the general health and well-being of children from same-sex parent families, compared with those from different-sex parent families.

Among both groups, a more positive parent-child relationship was linked to better general health and coping and learning behaviors, while a better partner relationship was associated with fewer emotional difficulties among children.

Same-sex parent families did report experiencing higher levels of stress than different-sex parent households, but the researchers note that this had no impact on child outcomes.

Prof. David A. Mills, PhD, who is the Shields Endowed Chair in Dairy Food Science, and colleagues had also discovered that the infant-associated subspecies of a particular bacterium, known as Bifidobacterium longum subsp. infantis, or B. infantis, produces a particular enzyme known as endo-β-N-acetylglucosaminidase (EndoBI-1).

EndoBI-1, which was identified by Prof. Mills’ team, can separate the oligosaccharides from the milk glycoproteins.

In the current study, Prof. Mills and coauthors hypothesized that these oligosaccharides were the food source for B. infantis.

The researchers have demonstrated that not only can the enzyme break down glycoproteins and release the oligosaccharides from the mother’s milk, but that it could do the same for cow’s milk.

They showed that released oligosaccharides are a substrate for the growth of B. infantis, but they also found that the oligosaccharides do not nourish adult-associated bifidobacteria.
